Introduction

 

This kit was designed for fixing various forms of biological materials, followed by embedding in wax or OCT. compound, for tissue section. Since cellular decomposition begins immediately after the death of an organism, biologists must fix the cells to prevent alterations in their structure through decomposition. Routine fixation involves the chemical cross-linking of proteins (to prevent enzyme action and digestion) and the removal of water to further denature the proteins of the cell. Heavy metals may also be used for their denaturing effect. Blocks of tissue (liver, kidney, pancreas¡K etc.) of approximate 1 cm 3 are rapidly removed from a freshly killed organism and placed in the fix buffer. If you study the location of molecules within cells, you have to immobilize the cells before labeling. Molecules that are not immobilized may be displaced from their normal location resulting in either a false localization or a negative result. The easiest method of immobilization is by chemical fixations, which have been traditionally used for this purpose. For immuno-cytochemistry tissues or cells can be fixed by immersion in a fix buffered solution.
